Amlodipine besylate and benazepril hydrochloride capsules are a combination antihypertensive available in six strengths (2.5/10, 5/10, 5/20, 5/40, 10/20, 10/40 mg) for oral administration.

Uses

Indicated for the treatment of hypertension in patients not adequately controlled on monotherapy with either agent.

Dosage

Initial dose is one capsule of 2.5 mg/10 mg once daily. Maximum dose is 10 mg/40 mg once daily. Antihypertensive effect largely attained within 2 weeks.

Side Effects

Most common reasons for discontinuation were cough (3.3%) and edema (2.1%). Other side effects include headache (2.2%), dizziness (1.3%), asthenia, flushing, dry mouth, nausea, and muscle cramps.

Interactions

Interactions with potassium supplements, lithium, NSAIDs, simvastatin (limit to 20 mg daily), mTOR inhibitors, and neprilysin inhibitors.

Warnings

BOXED WARNING: Fetal toxicity. Drugs that act directly on the renin-angiotensin system can cause injury and death to the developing fetus. Risks include angioedema, hypotension, hyperkalemia. Contraindicated with neprilysin inhibitors and in patients with angioedema history.

Pregnancy

Can cause fetal harm when administered to a pregnant woman. When pregnancy is detected, discontinue as soon as possible.

Storage

Store at 25 degrees C (77 degrees F); excursions permitted to 15 to 30 degrees C (59 to 86 degrees F). Protect from moisture.
