Dosage

Initial dose: 250 micrograms (1 mL) intramuscularly. Subsequent doses at 1.5 to 3.5 hour intervals. Maximum dose: 12 milligrams. For postpartum hemorrhage: single or multiple doses at 15-90 minute intervals; maximum 2 milligrams (8 doses).

Side Effects

Most frequent: vomiting (approximately two-thirds), diarrhea, nausea (one-third), temperature increase greater than 2 degrees F (one-eighth), flushing. Post-marketing: hypersensitivity reactions including anaphylaxis and angioedema.

Interactions

May augment activity of other oxytocic agents. Concomitant use with other oxytocic agents not recommended.

Warnings

Use only with strict adherence to recommended dosages. Must be used by medically trained personnel in hospitals with intensive care and acute surgical facilities. Contraindicated in hypersensitivity, acute pelvic inflammatory disease, active cardiac, pulmonary, renal, or hepatic disease. Animal studies show embryotoxicity in rats and rabbits.
