Uses
Indicated for treating Attention Deficit Hyperactivity Disorder (ADHD) and Narcolepsy. For ADHD, diagnosis requires hyperactive-impulsive or inattentive symptoms causing impairment before age 7, present in multiple settings.
Dosage
Amphetamines should be administered at the lowest effective dosage. For ADHD in children 3-5 years: start 2.5mg daily, increasing by 2.5mg weekly. Children 6+ years: start 5mg once or twice daily, increasing by 5mg weekly. Rarely exceed 40mg/day. First dose upon awakening.
Side Effects
Cardiovascular effects include palpitations, tachycardia, and elevated blood pressure. CNS effects encompass psychotic episodes, overstimulation, tremor, and tics. Gastrointestinal effects include dry mouth and anorexia. Serious skin rashes including Stevens-Johnson syndrome reported.
Interactions
MAOIs and CNS stimulants can cause hypertensive crisis. CYP2D6 inhibitors increase exposure risk. Serotonergic drugs increase serotonin syndrome risk. Acidifying agents reduce efficacy; alkalinizing agents increase blood levels. Tricyclic antidepressants may cause sustained concentration increases.

Sudden cardiac death risk, blood pressure/heart rate elevation, psychiatric adverse reactions, growth suppression, seizure threshold lowering, peripheral vasculopathy, serotonin syndrome risk. Contraindicated with advanced arteriosclerosis, symptomatic cardiovascular disease, hypertension, hyperthyroidism, and MAOI use.
Pregnancy
Amphetamines should be used during pregnancy only if the potential benefit justifies the potential risk to the fetus. No adequate controlled studies exist.
Storage
Store at 20 to 25 degrees C (68 to 77 degrees F) in tight, light-resistant containers.
