Uses

Indicated in the management of exogenous obesity as a short-term adjunct (a few weeks) in a regimen of weight reduction based on caloric restriction in patients with an initial BMI of 30 kg/m2 or higher who have not responded to appropriate weight reducing regimen alone.

Dosage

One extended-release 75 mg tablet daily, swallowed whole, in midmorning.

Side Effects

Cardiovascular: precordial pain, arrhythmia, tachycardia, elevation of blood pressure, palpitation. CNS: increase in convulsive episodes, psychotic episodes, dyskinesia, blurred vision, nervousness, restlessness, dizziness, insomnia. GI: vomiting, diarrhea, nausea, constipation. Other: urticaria, rash, impotence, bone marrow depression.

Interactions

Hypertension may result with MAO inhibitors. Combined use with other anorectic agents contraindicated. Antidiabetic requirements may be altered. Concurrent use with general anesthetics may result in arrhythmias. May interfere with antihypertensive drugs.

Warnings

Should not be used with other anorectic agents. The use of anorectic agents for longer than 3 months was associated with a 23-fold increase in the risk of developing pulmonary hypertension. May impair ability to operate machinery or drive. Prolonged use may induce dependence. Contraindicated in pulmonary hypertension, advanced arteriosclerosis, hyperthyroidism, glaucoma, severe hypertension, agitated states, history of drug abuse, or concurrent use with MAO inhibitors.

Pregnancy

Category B: Reproduction studies in rats at doses up to 1.6 times the human dose revealed no evidence of impaired fertility or harm to the fetus. Use only if clearly needed. Not recommended for nursing mothers.

Storage

Store at 20 to 25 degrees C (68 to 77 degrees F). Keep tightly closed and protect from excessive heat.
