Eslicarbazepine acetate is a dibenz[b,f]azepine-5-carboxamide derivative available as white to off-white tablets in strengths of 200 mg, 400 mg, 600 mg, and 800 mg.

Uses

Eslicarbazepine acetate tablets are indicated for treating partial-onset seizures in patients 4 years of age and older.

Dosage

Adult initial dose: 400 mg once daily, increasing weekly by 400-600 mg to maintenance of 800-1,600 mg daily. Pediatric dosing is weight-based. Reduce by 50% in moderate/severe renal impairment.

Side Effects

Most common: dizziness, somnolence, nausea, headache, diplopia, vomiting, fatigue, vertigo, ataxia, blurred vision, and tremor.

Interactions

Carbamazepine reduces eslicarbazepine levels; enzyme-inducing AEDs may require higher doses. Inhibits CYP2C19 and may induce CYP3A4. Reduces hormonal contraceptive effectiveness.

Warnings

Monitor for suicidal behavior, serious dermatologic reactions (SJS/TEN), DRESS, anaphylaxis, hyponatremia, neurological effects, liver injury, and hematologic abnormalities. Contraindicated in patients with hypersensitivity to eslicarbazepine acetate or oxcarbazepine.

Pregnancy

Animal studies showed developmental toxicity including malformations and fetal growth retardation. Limited human pregnancy data. Eslicarbazepine is present in human milk; effects on breastfed infants unknown.

Storage

Store at 20C to 25C (68F to 77F); excursions permitted 15C to 30C (59F to 86F).
